Contents Bevacizumab
Indications In combination w/ fluoropyrimidine-based chemotherapy for treatment of patients w/ metastatic carcinoma of the colon or rectum. In combination w/ standard cytotoxic chemotherapy for the 1st-line treatment of patients w/ locally recurrent or metastatic breast cancer (mBC). In combination w/ taxanes, capecitabine or gemcitabine for treatment of patients w/ mBC after failure of 1st-line cytotoxic chemotherapy. In addition to platinum-based chemotherapy for 1st-line treatment of patients w/ unresectable advanced, metastatic or recurrent non-squamous nonsmall cell lung cancer (NSCLC). In combination w/ interferon α-2a for 1st-line treatment of patients w/ advanced &/or metastatic renal cell cancer (mRCC). As a single agent or in combination w/ irinotecan for the treatment of glioblastoma after relapse or disease progression.

Dosage Metastatic colorectal cancer 1st-line treatment: 5 mg/kg body wt once every 2 wk or 7.5 mg/kg body wt once every 3 wk. 2nd-line treatment: 10 mg/kg body wt every 2 wk or 15 mg/kg body wt once every 3 wk. Continue treatment until progression of underlying disease. Metastatic breast cancer 10 mg/kg body wt once every 14 days or 15 mg/kg body wt given once every 21 days as IV infusion. Continue treatment until progression of underlying disease. Advanced, metastatic or recurrent NSCLC 7.5 mg/kg body wt every 3 wk in addition to cisplatin-based or 15 mg/kg body wt every 3 wk in addition to carboplatin-based for up to 6 cycles, followed by Avastin single agent until progression. Advanced &/or mRCC 10 mg/kg body wt every 2 wk until progression. Glioblastoma 10 mg/kg body wt once every 2 wk or 15 mg/kg body wt once every 3 wk.

Overdosage View Avastin overdosage for action to be taken in the event of an overdose.
Contraindications Hypersensitivity to Chinese hamster ovary cell products or other recombinant human or humanised antibodies.

Special Precautions Increased risk for development of GI perforation when treated w/ Avastin & chemotherapy. Increased incidence of HTN. Temporarily discontinue treatment in patients w/ HTN until adequately controlled. Discontinue use in cases of uncontrolled HTN, hypertensive crisis, development of arterial thromboembolic events & pulmonary embolism. Preexisting CAD or CHF. May adversely affect wound healing process. Initiate therapy only at least 28 days following major surgery or until surgical wound is fully healed. Pregnancy & lactation until 6 mth after last dose.

Adverse Drug Reactions GI perforations or fistulae, tumour-associated & mucocutaneous hemorrhage, arterial & venous thromboembolism, HTN, proteinuria, CHF, immune system disorders, hypersensitivity & infusion reactions.
