Treatment of HIV infection in combination w/ nucleoside analogues &/or PIs.

Dosage |
Combined w/ other PIs: 100-400 mg bid. Used as a single PI: 600 mg bid. Childn 400 mg bid. Max: 600 mg bid. Start at 250 mg/m2 & increase at 2-3 day intervals by 50 mg/m2 bid.

Contraindications |
Hypersensitivity. Co-administration w/ drugs highly dependent on CYP3A for clearance & for which elevated plasma conc are associated w/ serious events.

Adverse Drug Reactions |
Asthenia, GI & neurological disturbances including nausea, diarrhea, vomiting, anorexia, abdominal pain, taste perversion & circumoral & peripheral paresthesias.

Drug Interactions |
Decreased plasma conc w/ phenobarb, carbamazepine, dexamethasone, phenytoin, rifabutin & rifampicin. Increased plasma conc of certain drugs highly metabolized through CYP3A.

Pregnancy Category (US FDA) |
Category B: Either animal-reproduction studies have not demonstrated a foetal risk but there are no controlled studies in pregnant women or animal-reproduction studies have shown an adverse effect (other than a decrease in fertility) that was not confirmed in controlled studies in women in the 1st trimester (and there is no evidence of a risk in later trimesters). |
