性状
加巴喷丁是白色结晶粉末,以淡黄白色。在水或甲醇易微溶,乙醇(99.5),以微溶的,并在乙醚中几乎不溶。
适应病症
联合治疗抗癫痫药物对其它抗癫痫药物中充分的效果没有观察到癫痫患者的部分发作(有或没有继发性全身性发作)
用法用量
一般,分为口服给药成年和加巴喷丁至13岁的年龄和年龄较大的儿童的每日剂量为600mg,一日2日剂量1200毫克至每3倍的第一天。3天后,以口服给药分裂以每日剂量三次1200毫克-1800mg作为维持剂量。此外,虽然根据症状,每天和高达2400mg最高剂量调整。
通常,3至12岁的婴幼儿,以口服给药划分到三次的10毫克/千克,一日2日剂量为20mg/kg的加巴喷丁的每一天的日剂量。由于后3天的维持剂量,三至四岁的婴幼儿口服划分的每日剂量为25-35毫克/公斤三次在每日剂量为40mg/kg时,5-12岁的婴儿和儿童。可以根据症状,每天和高达50mg-/-kg的最高剂量调整。应当指出的是,相对于施用在一个时期的数量,并且不超过成人剂量和13岁或年龄较大的儿童。
药效药理
1. 休克惊厥模型抗惊厥作用
加巴喷丁,是通过在小鼠和大鼠的剂量依赖性方式的最大电击的强直性伸展痉挛。此外,加巴喷丁降低最大电击痉挛的抑制作用苯妥英钠的ED 50值,在卡马西平和丙戊酸的小鼠。
2. 在药物性模型抗惊厥作用
加巴喷丁,戊四氮,荷包牡丹,印防己毒素,士的宁和氨基硫脲诱发强直性伸展痉挛抑制。此外,也有人抑制戊四氮诱发的阵挛性惊厥,并没有打压荷包牡丹和印防己毒素诱发的阵挛性惊厥。
3. 在点燃模型抗惊厥作用
加巴喷丁,在海马点燃大鼠,以提高检动作,缩短了后燃烧的持续时间。
4. 在基因动物模型抗惊厥作用
加巴喷丁,抑制听源性癫痫发作和鼠标沙鼠的反射性癫痫。另一方面,它没有显示出对大鼠失神发作(小发作)和狒狒光原肌阵挛发作的效果
5. 作用机序
虽然加巴喷丁的抗惊厥作用的作用机制是未知的,加巴喷丁不结合的各种受体和主要的离子通道,包括从现有的抗癫痫药物的GABA相关受体,抗惊厥在不同的机制它已经建议行动的发作。
在附连到α2δ电压依赖性钙通道亚基的突触前抑制钙流入被认为有助于抑制兴奋性神经递质的释放。此外,已经发现,增加大脑GABA重量,它的贡献是未知。

Effects of this medicine
This medicine exerts anticonvulsant effect by two types of actions: decreasing calcium inflow and free neurotransmitter in the excitatory presynaptic terminal and increasing GABA in the brain to maintain/strengthen the GABA in nerve function.
Usually, used for treatment of partial seizures of epilepsy with concomitant therapy.
Before using this medicine, be sure to tell your doctor and pharmacist
•If you have previously experienced any allergic reactions (itch, rash etc.) to any medicines.
•If you have: renal impairment.
•If you are pregnant or breastfeeding.
•If you are taking any other medicinal products. (Some medicines may interact to enhance or diminish medicinal effects. Beware of over-the-counter medicines and dietary supplements as well as other prescription medicines.)
Dosing schedule (How to take this medicine)
•Your dosing schedule prescribed by your doctor is <<to be written by a healthcare professional>>
•For adults and children over 13 years old: in general, take 1 tablet (200mg of the active ingredient) 3 times on the first day. On the second day, take 2 tablets (400mg) 3 times. From the third day on, take 2 to 3 tablets (400 to 600mg) 3 times a day as maintenance doses. The dose may be adjusted according to your symptoms. The maximum daily dose is 12 tablets (2,400 mg).
For children 3 to 12 years old: in general, take 10mg/kg as the active ingredient in 3 divided doses on the first day. On the second day, take 20mg/kg in 3 divided doses. From the third day on, take 40mg/kg in 3 divided doses for children 3 to 4 years old and 25 to 35mg/kg in 3 divided doses for children 5 to 12 years old as maintenance doses. The dose may be adjusted according to your symptoms. The maximum daily dose is 50mg/kg. The dose in children 3 to 12 years old should not exceed the dose in adults and children over 13 years old.
Each tablet contains 200mg of the active ingredient. It may be combined with other antiseizure drugs. In each case, strictly follow the instructions.
•If you miss a dose, take the missed dose as soon as possible. The maximum time interval between doses should not exceed 12 hours. You should never take two doses at one time.
•If you accidentally take more than your prescribed dose, consult with your doctor or pharmacist.
•Do not stop taking the medicine unless your doctor instruct you to do so.
Precautions while taking this medicine
•The medicine may cause somnolence, or decrease of attention, concentration or reflex action. Avoid driving a car or operating dangerous machinery.
•The medicine may cause an increase of body weight. If you have signs of overweight, check with your doctor or pharmacist.
Possible adverse reactions to this medicine
The most commonly reported adverse reactions include somnolence, dizziness, headache, ocular disturbance such as diplopia and blurred vision, fatigue, convulsion, excessive appetite, excessive salivation and rash. If any of these symptoms occur, consult with your doctor or pharmacist.
The symptoms described below are rarely seen as initial symptoms of the adverse reactions indicated in brackets. If any of these symptoms occur, stop taking this medicine and see your doctor immediately.
•decreased urination output, swelling of hands/feet and face, fatigue [acute renal failure]
•high fever, reddened skin, stomatitis [Stevens-Johnson syndrome]
•rash, fever, lymph node enlargement [drug-induced hypersensitivity syndrome]
•general malaise, loss of appetite, yellowing of the skin or whites of the eyes [hepatitis, hepatic impairment, jaundice]
•muscle pain, feeling of weariness, reddish brown urine [rhabdomyolysis]
The above symptoms do not describe all the adverse reactions to this medicine. Consult with your doctor or pharmacist if you notice any symptoms of concern other than those listed above.
Storage conditions and other information
•Keep out of the reach of children. Store away from direct sunlight, heat and moisture.
•Since this medicine may be affected by heat, do not keep at high temperature and keep in a cool place.
•Discard the remainder. Do not store them.
