Ramelteon是一种结晶或结晶粉末的白色带淡黄色白色。 易溶于乙醇 (99.5) 和苯醇, 微溶于乙腈, 很难溶于水。
药用药理学
1. 作用机制
Ramelteon 是一种褪黑激素受体激动剂, 对褪黑激素MT1和MT2受体有较高的亲和力, 对人的褪黑激素MT1和mt2受体的亲和力(Ki值)分别为14.0pmol/L 和112pmol/L。在福斯科林诱导的cAMP生成反应指数中, 激动剂活性的IC 50值分别为21.2 pmol/L 和 53.4 pmol。Ramelteon是GABA, 包括GABAA受体, 血清素, 多巴胺, 神经递质受体, 如去甲肾上腺素和乙酰胆碱, 不表现出可检测的亲和力, 浓度为10[mu] mol只能。
2. 对睡眠的影响
口服0.03mg/kg对仙人掌夜间睡眠的影响显著降低了较浅NREM睡眠和慢波睡眠的潜伏期, 显著增加了总睡眠量。
在对猫睡眠觉醒的作用下, 通过口服拉梅特翁0.001mg kg缩短苏醒时间, 显著增加慢波睡眠时间。
适应症
失眠症患者睡眠困难的改善
用法与用量
通常情况下, 成人在睡觉时口服一次8毫克。

Indication
Rozerem is approved for adults having trouble falling asleep.
Important Safety Information If you have an allergic reaction, such as swelling of the tongue or throat, trouble breathing, and nausea and vomiting, contact your doctor immediately, as an airway obstruction due to this reaction could be fatal.
Don't take Rozerem if you're taking LUVOX® (fluvoxamine) or have severe liver problems. 
Talk with your healthcare provider if you have a history of depression, mental illness, or suicidal thoughts. Call your healthcare provider if your insomnia worsens or is not better within 7-10 days.
Worsening of depression, including suicidal thoughts and completed suicides, hallucinations, and nightmares can occur with Rozerem.
If you experience sleepwalking, driving, eating, or other behaviors while not fully awake, without remembering the event, contact your
doctor immediately.
There is a higher chance of these behaviors if you drink alcohol with Rozerem.
Don't drive or operate machinery until you know how you'll react to Rozerem.
Take Rozerem right before bed.
Do not drink alcohol or take other medicines that can make you sleepy when using Rozerem.
Rozerem may affect testosterone and prolactin. Consult your doctor if you experience changes in your period, libido, or problems with
fertility.
Side effects may include somnolence, dizziness, fatigue, nausea, and exacerbated insomnia.
