Contents Bevacizumab
Indications In combination w/ fluoropyrimidine-based chemotherapy for treatment of metastatic colorectal cancer (mCRC). In combination w/ paclitaxel for 1st-line treatment of metastatic breast cancer (mBC). In addition to platinum-based chemotherapy for 1st-line treatment of unresectable advanced, metastatic or recurrent non-squamous non-small cell lung cancer (NSCLC). In combination w/ interferon α-2a for 1st-line treatment of advanced &/or metastatic renal cell cancer (mRCC). As a single agent for treatment of glioblastoma w/ progressive disease following prior therapy.
Dosage mCRC 5 or 10 mg/kg body wt once every 2 wk or 7.5 or 15 mg/kg body wt once every 3 wk. mBC 10 mg/kg body wt once every 2 wk or 15 mg/kg body wt once every 3 wk. NSCLC 7.5 or 15 mg/kg body wt once every 3 wk in addition to platinum-based chemotherapy for up to 6 cycles, followed by Avastin Roche as single agent until disease progression. Advanced &/or mRCC 10 mg/kg body wt once every 2 wk. Glioblastoma 10 mg/kg body wt once every 2 wk. Doses are given as an IV infusion.
Contraindications Hypersensitivity to Chinese hamster ovary cell products or other recombinant human or humanised Ab. Patients w/ recent pulmonary haemorrhage/haemoptysis (>½ tsp red blood). Pregnancy & lactation.
Special Precautions Discontinue use in cases of GI perforation, tracheoesophageal fistula or any grade 4 fistula, grade 3 or 4 bleeding, uncontrolled HTN, hypertensive crisis, hypertensive encephalopathy, grade 4 pulmonary embolism, grade 4 proteinuria & development of reversible posterior leukoencephalopathy syndrome, arterial thromboembolic events. CV disease or CHF. May adversely affect wound healing process. Initiate therapy only at least 28 days following major surgery or until surgical wound is fully healed.
Adverse Drug Reactions GI perforations; pulmonary haemorrhage/haemoptysis; arterial thromboembolisms. HTN, fatigue or asthenia, diarrhoea, abdominal pain.
