Contents Simvastatin
Indications Listed in Dosage.
Dosage Reduction of total cholesterol conc in patients w/ primary hypercholesterolaemia, heterozygous familial hypercholesterolaemia & combined hyperlipidaemia when response to diet or other appropriate measures are inadequate 10 mg daily at night, adjusted at intervals of not <4 wk. Usual range: 10-80 mg once daily at night. Homozygous familial hypercholesterolaemia 40 mg daily at night or 80 mg daily in 3 divided doses (20 mg in 1st 2 doses & an evening dose of 40 mg). Prevention of coronary events & to slow progression of coronary atherosclerosis in patients w/ CHD & total cholesterol conc ≥5.5 mmol/L Initially 20 mg daily at night, adjusted at intervals of not <4 wk. Max: 80 mg daily.
Administration May be taken with or without food (Avoid excessive consumption (>1 L/day) of grapefruit juice.).
Contraindications Acute liver disease or unexplained persistently raised serum-aminotransferase conc. Pregnancy.
Special Precautions Monitor liver function periodically. Severe renal impairment. Discontinue use if marked or persistent increase in serum-aminotransferase or creatine phosphokinase conc occur.
Adverse Drug Reactions Digestive disturbances, muscle pain, tenderness or weakness, liver problems, hypersensitivity reactions.

Drug Interactions Ciclosporin, itraconazole, ketoconazole, erythromycin, clarithromycin, nefazodone, HIV-protease inhibitors, fibric acid derivatives, nicotinic acid. Coumarin anticoagulants.

The drug is contraindicated in women who are or may become pregnant.
