適應症 |
RA, heavy-metal poisoning. |
|
用量 |
Adult RA Initially 300 mg daily. Increase 300 mg in interval of 2 or 4 weeks till 600 mg daily. Max: 1.8 g. Maintenance: 300-600 mg daily. Acute & chronic heavy metal poisoning 900 mg-1.8 g daily depending on the severity of poisoning. Children RA Initially 5 mg/kg. Then increase 150-300 mg every 14 days till 15-20 mg/kg daily. May increase to 30 mg/kg. Usual dose: 5-10 mg/kg daily. |

用法 |
Should be taken on an empty stomach (Take on an empty stomach 1 hr before or 2 hr after meals, & at least 1 hr before or after any other food, milk, or medicine.). |

Category D: There is positive evidence of human foetal risk, but the benefits from use in pregnant women may be acceptable despite the risk (e.g., if the drug is needed in a life-threatening situation or for a serious disease for which safer drugs cannot be used or are ineffective). |

禁忌 |
Severe hematological dysfunction, severe renal failure, hypersensitivity to penicillin, SLE. Avoid in pregnancy & lactation. |

注意事項 |
Vit B6 supplement is required if high dose of penicillamine is given. Initiate hematological & urine monitoring 2 months before commencement of therapy. Monitor for psychological disturbance & blurred vision. |

不良反應 |
Skin reactions, exfoliative dermatitis, gastritis, taste perversion, renal insufficiency, hematological changes, neurological disturbances, immunological reactions, cholestasis, alopecia, breast enlargement.
