適應症 |
Treatment of adults with chronic, accelerated, myeloid or lymphoid blast phase chronic myeloid leukemia with resistance or intolerance to prior therapy including imatinib; adults with Philadelphia chromosome +ve acute lymphoblastic leukemia with resistance or intolerance to prior therapy. |

用量 |
Chronic phase CML 100 mg once daily. Accelerated phase CML, myeloid or lymphoid blast phase CML, Ph+ ALL 140 mg daily in 2 divided doses. |

過量 |
View Sprycel[柏萊] overdosage for action to be taken in the event of an overdose. |

用法 |
May be taken with or without food (Swallow whole, do not break/ crush/ cut.). |

注意事項 |
Complete blood counts should be performed weekly for the first 2 month & monthly thereafter or as clinically indicated. Caution to patients taking anticoagulants & antiplatelets. Patients who develop symptoms suggestive of pleural effusion eg dyspnea or dry cough should be eva luated by chest x-ray. Hypokalemia or hypomagnesemia should be corrected prior to administration. Patients who have/may develop prolonged cardiac ventricular repolarization (QT interval). Patients with congenital long QT syndrome. Patients taking arrhythmics or other medicinal products that lead to QT prolongation. Patients on cumulative high dose anthracycline therapy. Lactation, children & elderly. Hepatic & renal impairment. |

不良反應 |
Myelosuppression, bleeding related events, fluid retention, QT prolongation. Commonly reported: fluid retention eg pleural effusion. Diarrhea, headache, rash, nausea, hemorrhage, fatigue & dyspnea. Pyrexia, pneumonia, infection, febrile neutropenia, GI bleeding, dyspnea, sepsis, CHF.

交互作用 |
Strong CYP3A4 inducers (eg. dexamethasone, phenytoin, carbamazepine, rifampin, rifabutin, phenobarbital), St. John's wort. Strong CYP3A4 inhibitors (eg. ketoconazole, itraconazole, clarithromycin, atazanavir, indinavir, nefazodone, nelfinavir, ritonavir, saquinavir, telithromycin, voriconazole). Antacid, H2-blockers/proton-pump inhibitors (eg famotidine, omeprazole) & CYP3A4 substrates (eg alfentanil, astemizole, terfenadine, cisapride, cyclosporine, fentanyl, pimozide, quinidine, sirolimus, tacrolimus or ergot alkaloids).
