Indications |
Treatment of erectile dysfunction.

Dosage |
10 mg, approx 25-60 min before sexual activity. May be increased to 20 mg or decreased to 5 mg. Max: 20 mg/day. Max dosing frequency is once daily. A max dose of 5 mg should not be exceeded when used in combination w/ erythromycin. Elderly & patient w/ mild to moderate hepatic impairment or severe renal impairment Initially 5 mg, may be increased to 10 & 20 mg.

Contraindications |
Co-administration w/ nitrates or nitric acid donors. Men w/ severe CV disorders for whom sexual activity is inadvisable. Concomitant use w/ CYP3A4 inhibitors [ritonavir, indinavir, ketoconazole & itraconazole (oral)] in men >75 yr. Severe hepatic impairment, endstage renal disease requiring dialysis. Unstable angina. Known degenerative retinal disorders eg retinitis pigmentosa. Patients w/ anatomical deformation of the penis or conditions predisposing to priapism.

Special Precautions |
Concomitant use w/ ketoconazole, itraconazole, indinavir or ritonavir should be avoided. Avoid concomitant use w/ grapefruit juice. Symptomatic hypotension w/ α-blockers. Bleeding disorders or active peptic ulceration.

Adverse Drug Reactions |
Headache, flushing, dyspepsia, nausea, dizziness, rhinitis. Photosensitivity reaction, HTN. Rarely, visual disturbances.

Drug Interactions |
Plasma levels of vardenafil may increase when administered w/ erythromycin, ritonavir, indinavir & ketoconazole.

Pregnancy Category (US FDA) |
Category B: Either animal-reproduction studies have not demonstrated a foetal risk but there are no controlled studies in pregnant women or animal-reproduction studies have shown an adverse effect (other than a decrease in fertility) that was not confirmed in controlled studies in women in the 1st trimester (and there is no evidence of a risk in later trimesters). |
