Indications |
AIDS-related Kaposi's sarcoma in patients w/ low CD4 counts & extensive mucocutaneous or visceral disease. Ovarian & metastatic breast cancer, multiple myeloma.

Dosage |
AIDS-related Kaposi's sarcoma 20 mg/m2 IV infusion over 30 min every 2-3 wk. Ovarian/Metastatic breast cancer 50 mg/m2 IV infusion over 60 min every 4 wk. Multiple myeloma 30 mg/m2 on day 4 of bortezomib 3-wk regimen. Dilute in 250 mL (for doses <90 mg) or 500 mL (for doses ≥90 mg) dextrose 5% in water.

Contraindications |
Pregnancy & lactation. AIDS-KS that may be effectively treated w/ local therapy or systemic α-interferon.

History of CV disease, impaired cardiac function, patients who have received other anthracyclines, myelosuppression.

Adverse Drug Reactions |
Palmar-plantar erythrodysesthesia, leukopenia, anemia, thrombocytopenia, nausea, asthenia, alopecia, fever, diarrhea, infusion associated acute reactions, stomatitis, flushing, shortness of breath.

Drug Interactions |
In patients w/ AIDS-related Kaposi's sarcoma, potentiates the toxicity of cyclophosphamide & 6-mercaptopurine.

Pregnancy Category (US FDA) |
Category D: There is positive evidence of human foetal risk, but the benefits from use in pregnant women may be acceptable despite the risk (e.g., if the drug is needed in a life-threatening situation or for a serious disease for which safer drugs cannot be used or are ineffective). |
