Simvastatin |
|
Indications |
Treatment of CHD, hyperlipidemia. Adjunct to diet for reduction of elevated total & LDL cholesterol, triglyceride, & raises HDL. |

Dosage |
CHD Initially 10-20 mg once every evening. Hyperlipidemia Initially 10-40 mg once every evening. Dose Range 5-80 mg daily. Concomitant bile acid sequestrant, cyclosporine, danazol Initially 5 mg once daily. Maintenance: Max of 10 mg daily. Concomitant gemfibrozil Max: 10 mg daily. Severe renal insufficiency Initially 5 mg once daily with close monitoring. Max: 10 mg daily. |

Administration |
May be taken with or without food (Avoid excessive consumption (>1 L/day) of grapefruit juice.). |

Contraindications |
Active liver disease or unexplained persistent elevation of serum transaminases. Childn, pregnancy & lactation. |
|
Special Precautions |
Patient who consumes substantial amount of alcohol, history of liver disease. |
|
Adverse Drug Reactions |
Abdominal pain, flatulence, constipation, asthenia & headache. Rarely hepatitis, rhabdomyolysis.

Drug Interactions |
Anticoagulants, cyclosporin, fibric acid derivative, itraconazole, ketoconazole, erythromycin, nefazodone, immunosuppressant.

Pregnancy Category (US FDA) |
Category X: Studies in animals or human beings have demonstrated foetal abnormalities or there is evidence of foetal risk based on human experience or both, and the risk of the use of the drug in pregnant women clearly outweighs any possible benefit. The drug is contraindicated in women who are or may become pregnant. |
