Indications |
Breast cancer: Treatment of patients w/ 1st-line metastatic breast cancer who have tumors that overexpress human epidermal growth factor receptor 2 protein (HER2). Treatment of patients w/ HER2+ve early breast cancer (FISH+ or IHC3+) following surgery, chemotherapy & radiotherapy. In combination w/ an aromatase inhibitor for the treatment of patients w/ hormone receptor positive metastatic breast cancer. Advanced gastric cancer: In combination w/ capecitabine or IV 5-FU & a platinum agent for the treatment of patients w/ HER2 positive advanced adenocarcinoma of the stomach or gastroesophageal junction who have not received prior anticancer treatment for their metastatic disease.

Dosage |
Wkly schedule: Initial loading dose of 4 mg/kg as 90-min IV infusion, followed by 2 mg/kg wkly until progression of disease. Alternative 3-wkly schedule: Initial loading dose of 8 mg/kg, followed by 6 mg/kg 3 wk later, then 6 mg/kg repeated at 3-wkly intervals. Patients w/ early breast cancer should be treated for 1 yr or until disease recurrence.

Special Precautions |
Preexisting cardiac dysfunction especially in combination w/ anthracycline & cyclophosphamide. Pregnancy & lactation.

Adverse Drug Reactions |
Abdominal pain, accidental injury, asthenia, back pain, chest pain, chills, fever, flu syndrome, headache, infection, neck pain, vasodilation, GI disturbances, diarrhea, peripheral edema, edema, arthralgia, myalgia, anxiety, depression, dizziness, insomnia, paresthesia, somnolence, asthma, increased cough, dyspnea, epistaxis, lung disorders, pleural effusion, pharyngitis, rhinitis, sinusitis, pruritus, rash, cardiac toxicity, hepatic toxicity. Fever & chills (1st infusion).
