药理作用
本品为β受体阻滞剂，兼有适度的α受体阻滞作用，能降低血压，治疗心绞痛及心律失常。该药同时具有骨骼肌β2受体阻断作用，在MPTM诱发的震颤(猴)试验中，被证实有抗震颤作用。本品具有β-受体阻滞作用及适度的α-受体阻滞作用，其作用强度之比为8∶1。β-受体阻滞作用可抑制亢进的心功能，使心肌耗氧量减少，血压降低；β-受体阻滞作用又使外周血管阻力不致上升。
药动学
口服后2h血药浓度达峰值，半衰期为10h。血中及尿中主要代谢物为氨基甲酰基水解后的活性代谢物。
适应证
用于轻至中度原发性高血压、心绞痛、心动过速性心律失常；也用于特发性震颤（原发性震颤)。
禁忌证
1.有重度心动过缓，房室传导阻滞(Ⅱ度、Ⅲ度)，窦房传导阻滞的患者。
2.有糖尿病性酮症，代谢性酸中毒的患者。
3.有可能出现支气管哮喘，支气管痉挛的患者。
4.有心源性休克的患者。
5.有肺动脉高压所致右心衰竭的患者。
6.有充血性心衰竭的患者。
7.孕妇或有怀孕可能的妇女。
注意事项
1.慎用：特发性低血糖、长时间禁食、末梢循环障碍循环；肝肾功能障碍者。
2.服药期间禁止驾车。
3.停药时缓慢减退。
4.正在服用类似化合物的心绞痛患者须缓慢减量。手术前48h内不宜给药。
不良反应
1.严重不良反应：偶见心衰，房室传导阻滞，心动过缓。
2.一般不良反应：头痛倦怠、软便腹泻、腹部不适、腹痛、恶心呕吐、偶尔哮喘、皮肤过敏、皮疹、瘙痒、有时出现GOT、GPT、BUN及血糖升高。
3.不良反应发生率为8.9%，主要有心动过缓、心悸、头晕、乏力、抑郁、腹痛、恶心、食欲缺乏、稀便、转氨酶升高、雾视、泪液分泌减少、水肿、血尿素氮升高。罕见咳嗽、皮疹、肌肉痛及空腹血糖升高。
用法用量
1.原发性震颤：成人每次5mg，每天2次，疗效不充分时可增加用量，每次10mg，每天2次，但每天不能超过30mg。
2.高血压(轻度～中度)、心绞痛、心律失常：成人每次10mg，每天2次，疗效不充分时，可增至每天30mg。
药物相应作用
1.联合应用对交感神经系统有抑制作用的药物(如利血平)有时可出现过度抑制状态，应减量慎重应用。
2.联合应用降血糖药有时可增强降血糖作用，故应慎重应用。
3.联合应用钙拮抗剂有时可相互增强作用。
4.可乐定有可能增强停药后的回跳现象。
5.联合应用砒二丙胺，普鲁卡因胺，阿马林有时可出现心脏传导系统阻滞现象。
