Treatment of advanced renal cell carcinoma which has progressed on or after treatment w/ VEGF-targeted therapy.

Dosage |
10 mg once daily.

Overdosage |
View Afinitor overdosage for action to be taken in the event of an overdose. |
Administration |
May be taken with or without food (Take consistently either always w/ or always w/o meals. Swallow whole, do not chew/crush. Avoid grapefruit juice.). |
Contraindications |
Hypersensitivity to rapamycin derivatives. Severe hepatic impairment (Child-Pugh class C), galactose intolerance, Lapp lactase deficiency or glucose-galactose malabsorption.

Special Precautions |
New or worsening resp symptoms. Treat preexisting infection prior to treatment. Monitor renal function, fasting serum glucose level, complete blood count. Peri-surgical period. May affect ability to drive or operate machinery. Childn <18 yr. Pregnancy & lactation.

Adverse Drug Reactions |
Infections; blood count, glucose, cholesterol, triglycerides or liver function test abnormalities; anorexia, abnormal taste, pneumonitis, dyspnoea, epistaxis, cough, stomatitis, diarrhoea, mucosal inflammation, vomiting, nausea, rash, dry skin, pruritus, fatigue, asthenia, peripheral oedema.

Drug Interactions |
Ketoconazole, itraconazole, posaonazole, voriconazole, telithromycin, clarithromycin, nefazodone, ritonavir, atazanavir, saquinavir, darunavir, indinavir, nelfinavir. Rifampicin, carbamazepine, phenobarb, phenytoin, efavirenz, nevirapine. Erythromycin, verapamil, oral ciclosporin, fluconazole, diltiazem, amprenavir, fosamprenavir. Avoid use of live vaccines, St. John's wort & grapefruit juice.
