Indications |
Treatment of type 2 DM in adults, as 1st-line therapy irrespective of wt consideration, when dietary management & exercise alone does not result in adequate glycaemic control. Monotherapy or in combination w/ other oral antidiabetic agents, or w/ insulin.

Dosage |
Adult Initially 500 mg bd-tds, 850 mg bd-tds. Max: 3 g/day. Childn from 10 yr & adolescent Initially 500 mg daily, 850 mg daily. Max: 2 g/day. Should be given alone or w/ insulin.

Should be taken with food (Swallow whole, do not chew/crush.). |
Contraindications |
Diabetic ketoacidosis, diabetic pre-coma. Renal failure or renal dysfunction, dehydration, severe infection, shock, intravascular administration of iodinated contrast agents. Cardiac or resp failure, recent MI, hepatic insufficiency, acute alcohol intoxication, alcoholism. Lactation.

Special Precautions |
Lactic acidosis is rare & may occur in significant renal failure. Regular renal monitoring is needed. Suspend therapy during surgery & clinical investigations using IV iodinated contrast media.

Adverse Drug Reactions |
GI disturbances, metallic taste. Rarely, lactic acidosis, mild erythema.

Pregnancy Category (US FDA) |
Category B: Either animal-reproduction studies have not demonstrated a foetal risk but there are no controlled studies in pregnant women or animal-reproduction studies have shown an adverse effect (other than a decrease in fertility) that was not confirmed in controlled studies in women in the 1st trimester (and there is no evidence of a risk in later trimesters). |
