Indications |
Advanced soft tissue sarcoma after failure of anthracyclines & ifosfamide. |
|
Dosage |
Starting dose: 1.5 mg/m2 BSA IV infusion over 24 hr w/ a 3-wk interval between cycles. Premed w/ corticosteroids eg dexamethasone 20 mg IV 30 min before each infusion. |

Contraindications |
Active serious/uncontrolled infection. Lactation. Concomitant use w/ yellow fever vaccine. |

Special Precautions |
Hepatic or renal impairment (CrCl <30 mL/min), neutropenia & thrombocytopenia, rhabdomyolysis & severe CPK elevations (>10 x ULN), liver function test abnormalities, inj site reactions if administered through a peripheral venous line. Avoid concomitant use w/ alcohol. May impair ability to drive or operate machinery. Pregnancy. |

Adverse Drug Reactions |
Increased blood creatine phosphokinase, blood creatinine, blood alkaline phosphatase, AST/ALT & γ-glutamyltransferase. Decreased blood albumin. Neutropenia, thrombocytopenia, anemia, leukopenia, headache, hyperbilirubinemia, nausea, vomiting, diarrhea, constipation, fatigue/asthenia, anorexia, CPK elevations & rhabdomyolysis, alopecia.

Ketoconazole, fluconazole, ritonavir, clarithromycin; rifampicin, phenobarb, St. John's wort; cyclosporine, verapamil.
