Indications |
Female Anovulation including polycystic ovarian disease (PCOD) in women unresponsive to clomiphene citrate. Controlled ovarian hyperstimulation (COH) in medically-assisted reproduction programmes eg IVF/ET, GIFT & ICSI. Male Deficient spermatogenesis due to hypogonadotrophic hypogonadism. |
Dosage |
Slow IM/SC. Female Anovulation 50 IU/day for 7-14 days. COH 100-225 IU for at least the 1st 4 days. Maintenance: 73-375 IU for 6-12 days or longer. Male 450 IU/wk in 3 divided dosage of 150 iu concomitantly w/ HCG for at least 3-4 mth. |
Contraindications |
Tumours of the ovary, breast, uterus, testis, pituitary or hypothalamus; primary gonadal failure. Female: Pregnancy; undiagnosed vag bleeding; ovarian cysts or enlarged ovaries not related to PCOD; malformations of the reproductive organs incompatible w/ pregnancy; fibroid tumours of the uterus incompatible w/ pregnancy. |
Special Precautions |
Presence of uncontrolled non-gonadal endocrinopathies should be excluded. Hypersensitivity to streptomycin &/or neomycin. Female Unwanted ovarian hyperstimulation; risk for thrombosis; severe obesity or thrombophilia, increased risk of venous or arterial thrombo-embolic events; increased risk of multiple gestation & ectopic pregnancy. Early ultrasound is important. Male Elevated FSH levels is unresponsive to Puregon/hCG therapy. |
Adverse Drug Reactions |
Headache. Female Abdominal distension & pain; OHSS; pelvic pain; inj site reaction. Male Acne, rash, epididymal cyst; gynaecomastia; inj site induration.

Drug Interactions |
Concomitant use w/ clomiphene citrate may enhance follicular response. A higher dose may be needed after pituitary desensitisation induced by a GnRH agonist.
