Indications |
Adult: Replacement therapy w/ pronounced growth hormone deficiency (GHD). Childn: Growth disturbance due to insufficient secretion of growth hormone & growth disturbance associated w/ Turner syndrome or chronic renal insufficiency. Prader-Willi syndrome (PWS) for improvement of growth & body composition.

Dosage |
GHD Adult 0.2-0.5 mg/day. Patients w/ adult onset of GHD 0.15-0.3 mg/day. Childn 0.025-0.035 mg/kg/day or 0.7-1 mg/m2 BSA/day. PWS 0.035 mg/kg/day or 1 mg/m2 BSA/day. Turner syndrome & chronic renal insufficiency 0.045-0.05 mg/kg/ day or 1.4 mg/m2 BSA/day. Growth disturbance in short childn born small for gestational age 0.035 mg/kg/day or 1 mg/m2 BSA/day.

Overdosage |

Contraindications |
Evidence of tumour activity. Childn w/ closed epiphyses. Patient w/ acute critical illness suffering from complications following open heart surgery, abdominal surgery, multiple accidental trauma, acute resp failure or similar conditions.

Special Precautions |
Observe for evidence of glucose intolerance; DM. Perform thyroid function test after starting treatment. Intracranial HTN. Severe obesity, history of resp impairment or sleep apnoea, or unindentified resp infection. Discontinue at renal transplantation. Pregnancy. Adolescent.

Adverse Drug Reactions |
Ab formation. Adult: Paraesthesia, stiffness in the extremities, arthralgia, myalgia, peripheral oedema. Childn: Transient local skin reactions.

Pregnancy Category (US FDA) |
Category B: Either animal-reproduction studies have not demonstrated a foetal risk but there are no controlled studies in pregnant women or animal-reproduction studies have shown an adverse effect (other than a decrease in fertility) that was not confirmed in controlled studies in women in the 1st trimester (and there is no evidence of a risk in later trimesters). |
