Indications |
In combination w/ dexamethasone for the treatment of multiple myeloma patients who have received at least 1 prior therapy.

Dosage |
Recommended starting dose: 25 mg once daily on days 1-21 of repeated 28-day cycle w/ dexamethasone 40 mg once daily on days 1-4, 9-12 & 17-20 of each 28-day cycle for the 1st 4 cycles of therapy, thereafter dexamethasone 40 mg once daily on days 1-4 every 28-day cycle.

Administration |
May be taken with or without food (Swallow whole, do not break/chew/open. Take at the same time each day.). |
Contraindications |
Pregnancy & lactation, women of childbearing potential.

Special Precautions |
Perform complete blood cell count including WBC, platelet count, Hb & haematorcrit at baseline, wkly for the 1st 8 wk of treatment & mthly thereafter. Renal impairment. Monitor for thromboembolism, signs of bleeding (petechiae & epistaxes), thyroid function, risk of tumour lysis syndrome. Lactose intolerance. May impair ability to drive or operate machinery.

Adverse Drug Reactions |
Anaemia, neutropenia, thrombocytopenia, haemorrhagic disorder, insomnia, constipation, diarrhoea, nausea, increase & decrease in wt, rash, muscle cramp, muscle weakness, fatigue, asthenia, peripheral oedema.

Erythropoietic agents, other agents associated w/ increased risk of thrombosis (eg HRT), OC, warfarin, digoxin.

Pregnancy Category (US FDA) |
Category X: Studies in animals or human beings have demonstrated foetal abnormalities or there is evidence of foetal risk based on human experience or both, and the risk of the use of the drug in pregnant women clearly outweighs any possible benefit.
